Promiscuous Binding Identification

SPR can be used to screen large libraries of compounds or fragments to a set of targets. Many chemical libraries contain compounds that are exhibiting atypical behaviour when binding to the target protein. Promiscuous binders achieve enzyme inhibition using a nonspecific aggregation-type binding mechanism to proteins. These compounds are a source of false-positive hits in biochemical inhibition assays and ar recommended to be removed from screening hit lists. SPR represents a robust approach to identify these molecules early in the lead generation process. (A.M. Giannetti et al., J. Med. Chem.; 2008; 51(3); 574-580)
