Active Concentration Determination

Determining active-site concentration is valuable during drug discovery and development of therapeutic proteins. Almost all therapeutic proteins induce antibody responses in patients. Their serum-concentrations can be monitored by SPR. By knowing the concentration, the kinetic profile can be measured which in turn can help design properties of therapeutic drugs.

The method relies on changes in binding rates with varying flow rates under conditions of partial mass transport, and does not require standards of known concentrations, given that the molecular mass of the molecule of interest is known.
